In Vitro Cytotoxicity Evaluation of Highly Absorbent Foam Dressings Based on Silver Zirconium Phosphate via IC50 Value

چکیده
In this study, we, for the first time, tried to apply IC50 values (inhibitory concentration estimated to affect the endpoint in question by 50%) in the MTT colorimetric assay to investigate the cytotoxic effects of highly absorbent foam dressings based on silver zirconium phosphate, a newly nanobased matrix. Our results showed that silver released from dressings based on silver zirconium phosphate attributed mainly to highly cytotoxic to L929 cells cultured with MEM containing 10% fetal bovine serum. In addition, we have also compared the IC50 values among different dilutions of AgNO3 solution, silver based dressing extracts and material reference control (ZDEC) extracts using the optimized MTT assay, along with characterizing the silver content in the dressing extracts using atomic absorption spectroscopy. Results have shown that the IC50 values of AgNO3 solution, silver based dressing extracts and ZDEC extracts are 3.5 μg/mL, 3.8 μg/mL and 8.4%, respectively. And there exist some good agreements between qualitative and quantitative evaluation method as well. In conclusion, our study has led to the view that the IC50 value is a promising quantitative index for screening cytotoxicity with regard to silver based dressings.

A comparative study of the cytotoxicity of silver-based dressings in monolayer cell, tissue explant, and animal models.
Over the past decade, a variety of advanced silver-based dressings have been developed. There are considerable variations in the structure, composition, and silver content of these new preparations. In the present study, we examined five commercially available silver-based dressings (Acticoat, Aquacel Ag, Contreet Foam, PolyMem Silver, Urgotul SSD). We assessed their cytotoxicity in a monolayer...
متن کاملComparative Evaluation of Silver-Containing Antimicrobial Dressings on In Vitro and In Vivo Processes of Wound Healing
OBJECTIVES To compare the in vitro and in vivo effects of silver products on wound healing. METHODS Eight silver products were compared to determine: fibroblast function using fibroblast-populated collagen lattices (FPCLs), fibroblast viability using the Trypan Blue exclusion test, and fibroblast mitochondrial activity using the MTT [yellow tetrazolium salt; 3-(4,5-Dimethylthiazol-2-yl)-2,5-d...
متن کاملEvaluation of low-adherent antimicrobial dressings
Aims: To investigate the physical and antimicrobial properties of low-adherent, silver-containing gelling fibre and certain adhesive foam dressings in vitro. Methods: An in vitro model was used to quantitatively measure fibroblast adhesion to dressings in their dry and hydrated states. Microscopy techniques were used to visualise and quantify aspects of dressing composition and structure. Final...
متن کاملUse of a new silver barrier dressing, ALLEVYN◊ Ag in exuding chronic wounds
Recognising and managing wounds at risk of infection is vital in wound management. ALLEVYN Ag dressings have been designed to manage exudate in chronic wounds that are at risk of infection; are displaying signs of local infection; or where a suspected increase in bacterial colonisation is delaying healing. They combine an absorbent silver sulfadiazine containing hydrocellular foam layer, with a...
متن کاملWound dressings in diabetic foot disease.
Wound dressings represent a part of the management of diabetic foot ulceration. Ideally, dressings should alleviate symptoms, provide wound protection, and encourage healing. No single dressing fulfills all the requirements of a diabetic patient with an infected foot ulcer. Dressings research in this area is generally poor. However, each category of dressings has particular characteristics that...
